NEW! Animal Free Collagenase Grades Available!
Researchers interested in regenerative tissue applications and/or avoiding potential viral and prion risks from commonly used animal sourced fermentation media components now have Animal Free (AF) Collagenases produced by Worthington.
Types AFA, AFB ands AFC collagenase are derived from cultures grown in medium completely devoid of animal based components and designed for bioprocessing applications where introduction of potential animal derived pathogens must be prevented is now available from Worthington. Levels of secondary proteases are similar to Types 1 and 2 collagenase.
These new AF types are also available 0.22 micron filtered and lyophilized in 50mg vials for single use convenience.
Several lots of AF Collagenases are now available for sampling and evaluation in your specific application(s). Please contact Technical Service at 800.445.9603, e-mail: techservice@worthington-biochem.com or request samples on-line, Collagenase Sampling Program.
For more information.
Animal Free Recombinant (AF) DNase I Available!
Bovine pancreatic deoxyribonuclease I produced recombinantly in yeast, Pichia pastoris, to decrease levels of contaminating RNase and eliminate potential pathogens associated with animal based materials is now produced by Worthington.
Bovine pancreas is a rich source of RNase A which is often found in many commercial DNase preparations. Producing DNase I by recombinant means in an organism with much lower levels of endogenous RNase greatly facilitates purification of an enzyme with undetectable levels of RNase. The processes involved in the production and isolation of recombinant DNase I are completely devoid of animal based components which eliminates the possibility of introducing animal derived pathogens into bioprocessing procedures.
Recombinant DNase I is suitable for such applications as:
• Removing genomic DNA from RNA preparations prior to RT-PCR
• Degradation of DNA templates after transcription reactions
• Removing unwanted DNA from samples prior to Northern blotting
• Removing DNA during biopharma and bioprocessing procedures

Celase® GMP Collagenase Blend Available
Worthington Launches Celase® GMP an Avian and Mammalian Tissue-free Collagenase and Neutral Protease Enzyme Blend Produced Under GMP Guidelines
New enzyme grade is now available for use in a wide range of applications from initial animal research through clinical trials.
LAKEWOOD, NJ (April 5, 2016) – Worthington announced today at the Experimental Biology Conference in San Diego, the launch of the new Celase® GMP Enzyme product offering. Manufactured by Cytori Therapeutics, this product line is ideal for cell isolation studies for laboratories looking to facilitate a smooth transition from bench and animal research to downstream clinical applications.
Celase® GMP is a proprietary enzyme containing a unique blend of collagenase, neutral protease and buffer salts that are produced using avian and mammalian tissue-free raw materials, aseptically processed, sterile filtered and highly purified under GMP guidelines. A single, sterile, ready to use enzyme containing both collagenase and a neutral protease is ideal for a wide range of adipose stem cell, biomedical and bioprocessing applications.
“This new Celase® GMP product offering is a cost-effective option for laboratories looking for a high-quality GMP enzyme to support their pre-clinical research,” said James Zacka, vice president at Worthington. “We believe the sooner a researcher recognize a potential for a clinical application it is to their long term regulatory benefit to qualify and use a GMP or equivalent grade of enzyme”
Not all research applications required the use a GMP grade enzyme in early phase studies. However, the recent FDA guidance issued for tissue and cell products specifically cite that GMP grade reagents should be used in drug-type validated processes. Subsequently, both regenerative medicine researchers and clinicians are now looking for GMP quality products with a smoother regulatory approval process as the goal.
“The Cytori Celase® Enzyme is currently used in US FDA approved clinical trials. We are excited to offer a foundational and versatile enzyme for advancing adipose-based research programs from pre-clinical to clinical levels while eliminating the need to perform costly and time consuming bridging studies,” said Jason Streeter, quality engineer at Worthington.

Collagenase Product(s) Raw Material Certification
Although it does not affect the majority of research customers, please be aware that the FDA has recently issued a 'Risk of BSE in Collagenase Enzymes' notice to all FDA-regulated users of Collagenases. (5/23/07 FDA Collagenase Note: PDF Version) Worthington currently provides Collagenase to many such customers for which this issue is likely to arise. We recently drafted a document (Collagenase Products Raw Material Certification) in response to the FDA's letter to all regulated Collagenase user, available on our website. Please note that although Worthington is not currently a cGMP manufacturer of collagenase we operate under GMP guidelines and are ISO9001:2000 Certified. In addition and most importantly, Worthington has never used Brain Heart Infusion (BHI) media in our collagenase production process. Other low-risk animal sourced raw materials are of USDA or equivalent inspected and approved as suitable for human consumption. We have also been working on developing a completely Animal Origin Free (AOF) collagenase product and have raised the priority of this due to recent inquiries such as this. We hope to have AOF Collagenase evaluation material available for sampling within a few months. We would welcome the opportunity to provide evaluation samples as soon as they become available if interested. Please let us know if we can be of additional assistance or if any additional information is required by any of your customers. We look forward to continuing to provide collagenase to meet the technical and regulatory requirements of the market.
HEPATOCYTE ISOLATION SYSTEM AVAILABLE!
Optimized and Functionally Tested Enzyme System For Hepatocyte Isolation
The Worthington Hepatocyte Isolation System has been developed to provide researchers with a reliable, convenient, and consistent hepatocyte cell isolation system. By using the pre-optimized combination of enzymes contained in this kit, it is possible to minimize the lot-to-lot variation and improve the quality of the isolated hepatocytes.
In addition, Worthington use-tests each lot by isolating hepatocytes from adult rat to assure performance, reliability, and consistent yield of viable cells. Each kit includes single-use enzyme vials, balanced salt solution, buffer, culture media and an optimized protocol for five separate adult rat liver perfusions. The method is based on that described by Berry, M.N., modified by Seglen, P.O., and further optimized in conjunction with several collaborating researchers.

Collagenase Lot Selection Tool Available Online!
Worthington's 'Collagenase Lot Selection Tool' is available online at our website. This new feature was designed to help researchers select and evaluate current collagenase lots that match previous lots or desired activity profiles. Users may enter target values for collagenase, caseinase, clostripan, and tryptic activities or specify previous lot numbers.
Each value can be weighted based on the relative level of importance to the application. After the search for matches is completed, a ranked list of collagenase lots currently available is generated. The selected lots can then be sampled simply by using the built in link to the Free Collagenase Sampling Program.
As always, Worthington Customer and Technical Service personnel are available via phone at 800.445.9603/732.942.1660 and e-mail to assist with collagenase or any other products.
